In February 2017, it was announced that Wu had been set to star in Jon M. Chu's adaptation of Crazy Rich Asians, based off the bestselling novel of the same name by Kevin Kwan. Although she had initially auditioned for the role of Rachel in mid-2016, Constance could not accept due to a conflict with her work on the television series Fresh Off the Boat. However, Wu wrote to Chu explaining her connection with Rachel's character, and convinced him to push back the production schedule by four months.

Wu's character, Rachel, is an Asian-American economics professor at NYU who travels to Singapore with her boyfriend, Nick, to meet his wealthy family. Crazy Rich Asians was released in the United States and Canada on August 15, 2018 by Warner Bros., and is the first major Hollywood studio film to feature an all-Asian cast since 1993's The Joy Luck Club. Crazy Rich Asians was met with high critical praise, and became the number one film in the U.S. and Canada during its opening weekend.
