Dr. Kellie N. Smith Biography

Dr. Kellie N. Smith is a cancer immunologist, associate professor of oncology, and director of the FEST and TCR Immunogenomics Core Facility at the Johns Hopkins Bloomberg Kimmel Institute for Cancer Immunotherapy. Smith leads a laboratory focused on lung cancer immunology and immunotherapy. She completed her doctoral work at the University of Pittsburgh School of Medicine, concentrating on T cell immunology and immunotherapy. During her fellowship at Johns Hopkins, Smith worked under the mentorship of Dr. Drew M. Pardoll, identifying correlates of response to anti-PD-1 immunotherapy in patients with advanced non-small cell lung cancer.

Smith was part of a research team that contributed to foundational work in harnessing the immune system to target lung cancer, which has influenced the development of immunotherapy across multiple cancer types. Since joining the faculty, Smith has collaborated with clinicians at Johns Hopkins and other institutions on immunotherapy clinical trials aimed at improving treatment options, preventing disease recurrence, and understanding predictors of response in both early and advanced-stage disease. While much of her research centers on early-stage and resectable lung cancer, Smith also leads or participates in projects involving head and neck cancers, sinonasal carcinoma, Merkel cell carcinoma, mesothelioma, and esophageal cancer.

Smith's laboratory developed a diagnostic platform using a liquid biopsy approach, termed MANAFEST (mutation associated neoantigen functional expansion of specific T-cells), to detect and monitor anti-tumor T cells. This patent-pending technique is used in multiple clinical trials and at several research institutions and biotechnology companies to monitor responses to mutation-associated neoantigens, endogenous retroviruses, tumor-associated antigens, and viral antigens. Smith has also applied this platform with high-dimensional omics approaches to study the transcriptional programming of tumor-reactive T cells in cancer patients treated with checkpoint blockade. Her co-authored publication, "Neoadjuvant PD-1 Blockade in Resectable Lung Cancer," in the New England Journal of Medicine is among the most cited works in the field of cancer immunotherapy.

Smith has received several awards, including the LCFA/IASLC Lori Monroe Scholarships in Translational Lung Cancer Research, the Young Investigator Grant, and the American Lung Association's Pierre Massion Lung Cancer Discovery Award. These recognitions have supported her research in immunotherapy and contributed to clinical trial research that led to FDA approval of immunotherapy for early-stage lung cancers.

Smith serves on the Lung Cancer Foundation of America's Scientific Advisory Board. She has delivered keynote presentations on immunogenomic characterization of tumor-reactive T cells and has participated in public forums to communicate the science of immunotherapy to patients and broader audiences. Smith's work focuses on identifying which patients benefit most from immune-based treatments and developing new approaches to expand the effectiveness of these therapies in cancer care.
